چکیده
An oligomeric intermediate in the biosynthetic pathway of peptidoglycan was isolated from B. megaterium. The oligomer has been identified as [disaccharide(pentapeptide)]12 pyrophosphoryl undecaprenol.

Inhibition of peptidoglycan biosynthesis by ramoplanin.
Ramoplanin, a new lipoglycopeptide antibiotic, inhibits cell wall peptidoglycan biosynthesis in gram-positive bacteria. In both Staphylococcus aureus and Bacillus megaterium, UDP-N-acetylmuramyl-pentapeptides (UDP-MurNAc-pentapeptides) accumulated at concentrations of ramoplanin close to the MIC, indicating that inhibition of peptidoglycan biosynthesis occurred after formation of cytoplasmic pr...
متن کاملPurification of the peptidoglycan transglycosylase of Bacillus megaterium.
The peptidoglycan transglycosylase of Bacillus megaterium has been purified approximately 500-fold from a crude membrane fraction. This protein is likely to be the one previously called PG-II and was assayed by its ability to reconstitute with a crude phospho-N-acetyl-muramyl-pentapeptide translocase preparation and partially purified N-acetylglucosaminyl transferase to give peptidoglycan synth...
متن کاملPenicillin-sensitive transpeptidation during peptidoglycan biosynthesis in cell-free preparations from Bacillus megaterium. I. Incorporation of free diaminopimelic acid into peptidoglycan.
The effect of penicillins and cephalosporins on the various transpeptidation reactions presented in the preceding paper is discussed. Inhibition by cloxacillin and penicillin G of [Wldiaminopimelic acid incorporation during peptidoglycan synthesis appeared to be partially reversible after the antibiotic was destroyed by treatment with penicillinase. Inhibition of the cross-linking reaction by c...
متن کاملSynthesis of cross-linked peptidoglycan attached to previously formed cell wall by toluene-treated cells of Bacillus megaterium.
Cells of Bacillus megaterium can use the externally added precursors UDP-N-acetyl-D-glucosamine and UDP-N-acetylD mU!‘aIUyl L danyl D ghItamy1 mes o diaminopimelyl D alanyl-n-alanine to synthesize peptidoglycan if the bacteria were first treated with toluene. Toluene-treated bacteria could polymerize these precursors into peptidoglycan which was attached to previously existing cell wall by pept...
متن کاملThe cortical peptidoglycan from spores of Bacillus megaterium and Bacillus subtilis is not highly cross-linked.
Determination by amino acid analyses of the percentage of diaminopimelic acid in the spore cortex of Bacillus megaterium and Bacillus subtilis which is involved in interpeptide cross-links gave values of 31 to 37%. This finding supports the idea that the cortex volume could undergo significant changes in response to changes in pH or ionic strength and could thus play an active role in reducing ...
